Abstract

Non-steroidal anti-inflammatory drugs (NSAIDs) are a class of medicines usedworldwide; they have low solubility, low bioavailability and adverse reactions. Thecarrying of these drugs provides a change in the pharmacokinetics and biodistributionof the drug, providing greater stability and reduction of adverse reactions. As promisingcarriers, Layered double hydroxides (LDH) are presented with wide versatility andproviding stability to the drug. However, understanding the interaction between the drugand the inorganic matrix (LDH), is extremely important, once the drug(s) may sufferchemical changes, influencing the compatibility/stability of the product, of the interactionbetween the inorganic matrix and the drug. The present work aimed to synthesize LDHof Mg/Fe and Mg/Al by the coprecipitation method, for the interaction with ibuprofen(IBU), as a model drug, to study the carrier and reactivity conditions, through theinvestigation of solid mixtures, and carrying out the incorporation by the ion exchangemethod, through mixtures in solution using different solvents (water, dimethylsulfoxide,acetone and chloroform), and different reaction pH. The characterizations of thesynthesized compounds were performed by infrared vibrational spectroscopy (IR);scanning electron microscopy (SEM) coupled with energy-dispersive X-rayspectrometer (EDX); and by X-ray diffraction (XRD) analysis. Desorption assays of theincorporation and dissolution hybrids of the formed complex were monitored bymolecular electron spectroscopy in the ultraviolet/visible (UV-Vis) region in phosphatebuffer solution (pH 7,4). it was observed that in solid mixtures there is a reaction betweenLDH and IBU with reactivity interaction. In aqueous medium, depending on the pH usedand the different solvents, the incorporation or reactivity of IBU with the matrix occurs.The reaction between LDH and IBU in aqueous medium with pH 9,0 control, and thereaction in acetone, showed a carrying capacity of around 27%, the reactions under thesame conditions LDH Mg/Al, no evidences of carrying was identified. In aqueousmedium without pH control and with pH 7.0 control, the reactivity between LDH Mg/Fewith IBU and LDH Mg/Al with IBU was confirmed, as evidenced by the monitoring of thestrong bands around 1540–1580 cm -1 and 1405–1440 cm-1, as identified in the literaturefor complexes with IBU. From the results, it is concluded that the incorporation by theion exchange method depends on the synthesis route (pH and solvent) and the carryingcapacity resembles the recurrent study. And the complex formation between the metal(III) and IBU is confirmed by the reaction between LDH and IBU, making it possible topropose a comparative structure with those in the literature for the new product formedin this work, opening up to important perspectives in the understanding of theinteractions between the IBU and matrices that carry intercalation or reactivity and inobtaining a new formulation.
